Purified urine exosomes (small EVs) are obtained from a pool of certified healthy donors. Exosomes/small EVs (40–120 nm) are purified using a double tangential flow filtration process using HBM-LS propreitary filters TFF-MV and TFF-EVs. Purified EVs are validated and characterized for overall protein content, marker expression (CD9, CD63, CD81), particle concentration and size distribution (Nanoparticle Tracking Analysis performed with the ZetaView® Evolution analyzer, Particle Metrix), and morphology (TEM).
The effects of lyophilization on EV protein stability are comparable to other storage methods, including storage at −80 °C. Lyophilized vesicles are easy to store and ship, and remain stable long-term for up to 36 months.
Applications: Control (spike-in) for EV quantification, protein marker analysis (ELISA, WB, flow cytometry). Extraction and analysis of EV associated nucleic acids (qPCR, ddPCR, RNA seq). NTA, fluorescent NTA (F-NTA). Single EV analysis by Electron microscopy (TEM), Fluorescent microscopy.
